2.These are my favourite blue jeans.

Which one is right? Why?

The second one. For some reason we use the plural form even though it is one item of clothing.
 
Matthew, dictionaries get their definitions from usage, not the other way around.
 
This is my favourite pair of jeans would work.
 
Yes, that's the whole point. Anything that is expressed as coming in pairs is, by definition, plural. Scissors and glasses are two more examples.
