1) Will Tom's restaurant on 23rd street be open?
2) Will Tom's restaurant, which is on 23rd street, be open?

Let's say that we know who Tom is and he has only one restaurant and it is on the 23rd street. Is #1 acceptable?
Could it be used instead of #2?
Does 'on 23rd street' need to be set off with commas in #1, as it is presenting non-essential information?

Let's say 'Foodjoint' is the name of a restaurant that does not belong to a chain. There is only one restaurant named 'Foodjoint' and 'Foodjoint' is a proper noun. Can #3 be used in this context?
Could it be used instead of #4?

"Will Tom's restaurant on 23rd street be open?" tells me that you are asking about that particular location and that there are other restaurants owned by that person.

Do you find that sentence acceptable if the meaning is that Tom has only one restaurant?

One, I wouldn't take that to mean Tom has only one restaurant. Two, you seem to be going out of your way to compose unnatural sentences. (See below.)

Abe: Is Tom's restaurant open?
Bob: Which one?
Abe: The one on 23rd Street.
Bob: That one? No, it's closed for good and has been for months. There's a closer one though.
