Hello.
Should I use ''at'' or ''in'' before ''wholesale'' and ''retail?''

For example, ''They prefer to buy items at/in wholesale, at/in retail.''
 
You can say "at wholesale" with the understanding that the word "prices" is being omitted. Or, just use "buy wholesale."
 
Neither needs a preposition of any kind when used as adverbs. The phrases are buy/sell something wholesale/retail.
