When you want to tell the person on the phone that the person they want to speak to cannot come the the phone, the correct question is 'Can I take a message?' but not 'Can I give a message to?' Am I right?

Yes. Or you could say:

XXX is busy at the moment. Would you like to leave him/her a message? Can I ask him/her to return your call?
