Is this sentence correct?

At 12 o’clock, we have Break – it’s lunchtime and a chance to catch up with all my mates on the latest news.

Yup!

A minor question: Should Break be a proper noun? When there's a choice, I usually avoid capitals. But it's your call.

Yes, thank you, I doubted about the capital letter. Do we need an article before break then? And what about the sentence itself - is it correct (now that I've changed it)?

At 12 o’clock, we have a break – it’s lunchtime and a chance to catch up with all my mates on the latest news.

Can 'on' be replaced with 'about' in the OP's sentence?

Yes, but 'on' is more natural.

Do we need an article before break then?

In most cases, it would sound better to me with the article a. If it's someone talking about the mid-morning break in school, then it would be OK without in BrE.
