A once actor (Yao) now is doing livestream on Chinese Tiktok; he’s teaching English there. Recently I watched a TV series and I saw a character looks really like Yao, so I went to Yao’s livestream (a chat room on TikTok) and asked him the question “Do you play a character in that series?” I mentioned the TV series’ name to him and wanted to make sure if he did play in that TV series.

Is my italic sentence natural?

See above.

You said he used to be an actor so you need to start your question with "Did you" - if he played that character it must have been in the past because he's no longer an actor.
