Absolutely there's a difference—a big difference! That's the whole point.
 
Well, I've got it. It's some dirty joke I don't clearly understand because of my poor English. I've got intrigued, I must confess, and looked up 'jack off' as a phrasal verb in the dictionary and found the meaning 'masturbate'. Is this the meaning?
 
The clue was in the lack of capital letters for the second 'uncle jack'.
