2. In a week's time I will have completed my work.

I would say both, as long as you add a comma after time. The perfect form adds an extra element of completion.

They mean different things. 2 means that your work will be complete in one week's time. 1 could mean (and usually does) that in a week's time, you will sit down and finish it. It implies that you will not work on it within the next week.
