What would be the correct option inside a text: ",...in spite of..." or "...despite of...". For example: "there is a large number of criticism is spite/despite of....."
 
Despite means In spite of. There is no such thing as despite of.
 
Yes, sorry, I meant ",...in spite of..." or "...despite...". Are both correct?
 
They are usually interchangeable, but your example 'sentence' is neither grammatical nor complete.
 
And it should be something like there's a lot of criticism, not number.
