Which one sounds natural in the sentence? And is there any difference in meaning?

1- Most people in the US/ Most of the people in the US are very rich and educated.

The shorter one is the more natural for me.

Note my corrections above. I've said it before and I'll say it again - as a learner, don't try to start sentences with "and" or "but". Don't leave a space after a slash mark.
 
They're both equally natural. They mean different slightly things.

  • To talk about a majority of a general group, use most + noun
  • To talk about a majority of a specific set of something, use most of the

In this case, using most of the people could sound like you are contrasting people with animals, for example.
 
emsr2d2 has a point, and so does jutfrank.
If you were talking about people in general, then say "Most people in the US". However, if you're talking about some specific group of people, then "Most of the people in the US" would be appropriate. For example:
"The firm surveyed 7,000 individuals in ten countries, and according to the results of the survey, most of the people in the US are very rich and highly educated".

All things are relative. Compared to people in some countries Americans are very well off indeed. There are, of course, some countries with a higher average annual income than ours. It depends on who is being compared to who.
