Which of the above- mentioned sentences is correct?
 
1. The medicine should be taken twice daily after meal for five days.
2. The medicine should be taken twice daily after meals for five days.

Which of the above-mentioned sentences is correct?
Only sentence #2 is correct.
Don't put a space after a hyphen. It would be more natural to ask "Which of the sentences above is correct?"
 
Only #2 works in AmE. I was about to post stating #1 was incorrect.
 
*Apologies for any confusion. I was sure I'd written "#2" in my response but I'd mistyped it as "#1". I've corrected it. To be clear, as Skrej said, #1 is incorrect.
