Is this sentence OK?

I just realized there was a typo in my earlier letter therefore I have prepared a new letter.

It is a run on sentence, i.e. two sentences improperly joined into one. "Therefore " does not work as a connective.

One solution would be to separate it into its two statements. "I made an error" is the first statement. "I did something about it" is the second statement. Each statement requires its own sentence.

Or you could combine them into a compound sentence using a correct connective such as but. "I made a mistake, but I ..."

Yes, that could be an example. However it does not sound as natural as other options could (It sounds more formal than the previous sentence.) (It is also a synonym of 'therefore'.)

"I had an error" is far less likely than "I made a mistake/error / There was an error". 'Thus' is not appropriate. It is better to say 'new letter' than 'another one' which could be interpreted as 'another mistake'.

I would personally have no problem with:
"I realise(d) (that) there was a typo in my earlier letter and so have prepared a new one."
