Dear Ms/Mr.
Hello,
I wanna know if one use this expression at the begining of his letter,
what shoul I reply? "Dear Ms/Mr."
i mean i should mention that i am female(Ms) in my response to his letter?
and another question is that how i should reply one's apology in a very formal letter?

In AmE, if you don't know the gender of the person who might be reading your letter, the traditional salutation is: "Dear Sir/Madam".
As for your other questions, we need more context. What type of letter are you sending? Why would you need to indentify your gender? etc

I just want to know is it necessary to indentify my gender in response to this expresson(Dear Ms/Mr.) ? without regarding the type of my letter.
my letter is an ordinary formal letter,actually a scientific letter.
