Which is correct?

Superlative needs a the.

Not a teacher. The first sentence is correct to me. Do you mean that this particular person is attractive all day long, but at night she is at her prettiest?

Then use the first sentence.

I would only use "the prettiest" if she was being compared to other people. "She is the prettiest of the girls in the rooms."

She is the prettiest (of all girls).
She is prettiest (at a specific place, time etc.).
