Are both sentences acceptable? Thanks.
 
I am not sure what you mean. Entering and exiting the bus? Walking up and down the aisle?

No, we don't say that.
 
We really only say "I get on the bus" or "I board the bus" and "I get off the bus".
Once inside the bus, we move back/along the bus to make room for people at the front. On a double decker bus, we go "upstairs".
