Q. My city name is Karachi. What do you call the residents of this city, either Karachities, Karachean or something else? What is the rule in English in this respect?

I think that we go for the one that sounds best and is easily recognisable, so I would use the same as Raymott.
 
And what are the people called who live in flats (irrespective of tenancy or land lordship)?

I'd call them "renters."
