Do we we say :"Their love and affection makes them want take care of him" or "Their love and affection make them want take care of him"

Do we we say no colon here "Their greed and cupidity makes them want more" or "Their greed and cupidity make them want more"?

I am certain that "cupidity" is not the word you meant to write. Please click on "Edit" and change it to the correct word.

Note my markups above. In future, there is no need for a greeting, and please don't say "Thanks" before anyone has done anything.
 
Technically, it should be "make". Some people might view "love and affection" as a single thing and use "makes".
 
@Wave_01 Please note that I have changed your thread title. "Grammar question" is not an appropriate or effective title. Titles should include some/all of the words/phrases/sentences you're asking us about.
 
This is the pattern:

Their X and Y make them Z.
