Which one is correct?

If you want to make a single sentence, write:

Is "You both have a good sense of humour" or "You both have good sense of humour" correct?

Note that when a quoted sentence forms part of a longer sentence, you don't include the full stop. However, that's the only punctuation mark that should be omitted. If a quoted question forms part of a longer sentence, you must include the question mark. For example:

Is "What's your name?" or "What your name is?" correct?

What I'm trying to get at is that the two full stops inside the quotes in your original layout were incorrect.
